How does Micro Spectrophotometer detect protein A280

Protein and nucleic acid are not the same, has the very strong diversity. Protein A280 function is used to detect pure proteins containing Trp, Tyr residues or Cys-Cys disulfide bonds, which are visible at 280nm. The native method does not need to construct a standard curve, but directly calculates the protein concentration after measuring the absorbance.
Protein A280 displays the UV absorption spectrum and calculates the concentration (mg/ mL) after detecting the absorbance at 280nm. As with nucleic acid assays, Protein A280 records display data at a 10mm optical path.
The Nano-600 can detect up to 90mg/ mL of BSA in pedestal mode without dilution.
When the light intensity after absorption of the test sample is less than 200 (under 10mm optical path), the software will prompt the user to choose a smaller measurement optical path to ensure the accuracy of the test. The screen is shown below.
The main factor that determines the surface tension of a liquid is the hydrogen bond between water molecules in the solution. Normally, substances in water, such as proteins, salt ions, detergents, etc. will reduce the surface tension by breaking the hydrogen bond between water molecules. Although 1ul is sufficient for most samples, 2ul is recommended to ensure column formation due to reduced surface tension.
Baseline calibration can be selected or cancelled. The default baseline calibration wavelength for protein detection is 340nm. Users can input different calibration wavelengths according to test needs. In any case, the baseline is automatically set to the absorbance at the selected wavelength, and the absorbance readings for all wavelengths are subtracted from this value.
Note: Baseline calibration must be set before sample testing is performed. Setting after sample testing is invalid. Without selecting the baseline calibration, the spectral values will be offset and the calculated concentrations will change.
